The Nuclear and Cytoplasmic Protein Extraction Kit offers a simple and convenient method for isolating nuclear and cytoplasmic proteins from cultured cells or fresh tissues. The separation process for nuclear and cytoplasmic proteins from cultured cells can be completed in approximately 90 minutes.
This kit uses Cytoplasmic Protein Extraction Reagents A and B to fully swell cells under low-osmotic pressure conditions, disrupt the cell membrane, and release cytoplasmic proteins. The nuclear fraction is then collected by centrifugation. Nuclear proteins are subsequently extracted using high-salt nuclear protein extraction reagents.
Additionally, the kit includes unique components for extracting membrane proteins from cells and tissues. Its specialized buffer system selectively separates and isolates cell membrane and organelle membrane proteins during cell lysis. The extraction process is simple, reliable, and rapid. The resulting membrane proteins are of high purity and are suitable for downstream applications such as SDS-PAGE, Western blotting, and immunoprecipitation.
Each extraction can process approximately 10⁷ cultured cells or 200 mg of animal tissue.
Application
The extracted proteins can be used for subsequent applications such as Western blotting, EMSA, footprinting, reporter gene assays, and enzyme activity assays.
