Background:
Histone threonine phosphorylation is a reversible modification regulated by kinases and phosphatases. Phosphorylation changes chromatin charge, recruits effector proteins to coordinate cell cycle progression, mitosis and DNA damage response, each threonine site carries unique functions. H3T6 phosphorylation (H3pT6) is catalyzed by cell cycle-related kinases, highly induced during mitosis. It loosens condensed mitotic chromatin and mediates spindle protein recruitment for chromosome segregation. This mark also accumulates at DNA double-strand break sites to activate repair signaling. H3T11 phosphorylation (H3pT11) responds to metabolic and mitotic signals. In proliferating cells, it accumulates at gene promoters to drive transcription; during mitosis, it participates in chromosome condensation and spindle assembly checkpoint control. Abnormal H3pT11 level leads to mitotic defects and genome instability.
