Background:Histone H3 forms H3-H4 tetramers as nucleosome skeleton. Lysine acetylation regulated by HAT/HDAC relaxes chromatin and recruits transcription co-activators. Different acetyl sites control transcription, DNA repair and chromatin assembly.H3K57 sits inside nucleosome core and acetylated by TIP60. H3K57ac weakens intranucleosome interaction to facilitate post-replication chromatin assembly and homologous recombination repair. Deficiency causes genome instability and high sensitivity to DNA damage agents.
