U-Blot® Histone H3 (Phospho Ser10/Thr11) Rabbit pAb
Product details
Background:Histones are basic nuclear proteins that assemble into nucleosomes, the fundamental repeating units of eukaryotic chromatin. The N‑terminal tail of histone H3 undergoes combinatorial post‑translational modifications. H3S10pT11p represents the dual phosphorylation mark on histone H3 at Serine 10 and Threonine 11. This combinatorial modification is predominantly deposited by Aurora‑B kinase during mitosis. Co‑phosphorylation of H3S10 and H3T11 occurs dynamically along mitotic chromosomes and peaks in M‑phase. Dual phosphorylation reinforces chromatin structural rearrangement, facilitates HP1 dissociation from condensed chromosomes, and contributes to sister‑chromatid resolution, proper chromosome compaction and accurate chromosome segregation. This double mark distinguishes mitotic chromatin from interphase chromatin carrying only single‑site H3S10 phosphorylation. Crosstalk exists between this dual phosphorylation and nearby lysine acetylation or methylation, orchestrating chromatin‑reader protein binding. Defects in H3S10pT11p deposition lead to mitotic malfunction, chromosome mis‑segregation and genomic instability, which are frequently observed in malignant cells.
