Background:Histone serine phosphorylation is reversibly regulated by kinases and phosphatases. Phosphorylation changes chromatin charge and recruits functional proteins to control histone deposition, DNA repair and cell cycle, with unique functions for each site. H4 Ser1 phosphorylation (H4pS1) is a conserved N-terminal modification of newly synthesized histone H4 after DNA replication. It mediates nucleosome assembly and stabilizes chromatin structure. This modification accumulates at DNA double-strand break loci to relax chromatin and assist the recruitment of DNA repair complexes.
