Product details
The BCA Protein Assay Kit is based on the BCA method, one of the two most commonly used protein concentration determination techniques.
Principle of the BCA Method: Under alkaline conditions, the peptide bonds in proteins bind with Cu²⁺, reducing it to Cu⁺. The BCA reagent specifically binds to Cu⁺, forming a stable blue-purple complex with a maximum absorbance of 562 nm. The intensity of the color is directly proportional to the protein concentration.
This assay offers simplicity, high stability, sensitivity, and compatibility in protein concentration determination.
The BCA method is compatible with most chemicals in samples, including up to 5% SDS, 5% Triton X-100, and 5% Tween 20, 60, or 80. However, it is sensitive to chelating agents and higher concentrations of reducing agents. Ensure the following conditions for accurate results:
- EDTA < 10 mM
- No EGTA
- Dithiothreitol < 1 mM
- β-mercaptoethanol < 0.01%
If the BCA method is not suitable, the Bradford Protein Assay Kit (BPK-1500) is recommended as an alternative.
Storage and stability:
BCA reagents A and B should be stored at room temperature, and protein standards should be frozen at -20℃. The validity period is one year.
