Background:
Histone H3.3 is a unique replication-independent H3 variant, distinct from canonical H3.1/H3.2 in amino acid sequence and chromatin deposition pattern. It is continuously expressed throughout the cell cycle and enriched in active transcription regions, enhancers, telomeres and DNA damage sites, mainly responsible for maintaining chromatin dynamic remodeling, transcriptional activity and genome stability. H3.3 plays a vital role in embryonic development, stem cell differentiation and immune regulation, and its mutation and dysregulation are closely related to multiple tumors.
