Background:Histones are basic nuclear proteins that assemble into nucleosomes, the fundamental repeating units of eukaryotic chromatin. Multiple post‑translational modifications can coexist on the same histone H3 N‑terminal tail, forming combinatorial epigenetic signals. H3K27me3S28p represents the dual modification of trimethyl‑Lysine 27 and phosphorylated‑Serine 28 on histone H3. H3K27me3 is deposited by Polycomb Repressive Complex 2 (PRC2), marking facultative heterochromatin and mediating developmental gene silencing. During mitosis, Aurora‑B kinase catalyzes phosphorylation at Ser28 on H3K27me3‑bearing histones.
