SDS-PAGE Protein Sample Loading Buffer (5X, Odorless) is an improved, safer, and healthier version of the conventional 5X protein loading buffer, using bromophenol blue as the dye. It replaces the odorous dithiothreitol (DTT) or mercaptoethanol (2-Mercaptoethanol) with a safer, more water-soluble reducing agent that offers similar reducing power. This ensures the loading buffer remains odorless during both normal use and heating, making protein loading safer and healthier.
Despite being odorless, this product performs the same as conventional protein loading buffers, showing no significant differences in SDS-PAGE protein sample loading.
Application
Used for routine SDS-PAGE protein sample loading.
Storage and stability:
Store in a sealed and frozen state at -20℃ away from light and in a dry place. The shelf life is 1 year.
Instructions for use
- Dissolve the SDS-PAGE loading buffer (5X) at room temperature or in a water bath not exceeding 37° Store at room temperature immediately after dissolving and avoid prolonged exposure to the water bath.
- Add 1 μL of protein loading buffer (5X) to every 4 μL of protein sample and mix.
- Heat at 100°C or in a boiling water bath for 3-5 minutes to fully denature the protein.
Note: If the initial amount of cells or tissues is large and the genomic DNA content is high, the sample may remain viscous after boiling. In this case, continue boiling for another 5-10 minutes, or dilute the protein loading buffer to 1X and then boil for an additional 3-5 minutes. This will release proteins from genomic DNA and reduce viscosity without affecting subsequent loading.
- After cooling to room temperature, load the sample into the SDS-PAGE gel.
- Electrophoresis can be stopped when the blue dye reaches the bottom of the gel.
