The BCIP/NBT Alkaline Phosphatase Substrate Solution produces a dark blue, insoluble end product and is used in immunohistochemistry as well as Northern and Southern hybridization involving alkaline phosphatase probes. 5-Bromo-4-chloro-3-indolyl phosphate (BCIP) and nitro blue tetrazolium chloride (NBT) are employed for the colorimetric detection of alkaline phosphatase-labeled molecules. BCIP is available in two salt forms, which can be used to prepare storage solutions and reaction buffers in combination with NBT to create the substrate solution for alkaline phosphatase. When incubated with alkaline phosphatase, the substrate system generates an insoluble purple NBT diformazan product that is easily visualized.
Application
Northern, Southern, and Western blotting, in situ hybridization, and immunohistochemistry—this reagent can be used for alkaline phosphatase-based color development in cells or tissues. It is also suitable for enzymatic color development in Western blot experiments.
